If I update a field after the sequence message has already been drafted, will it also update that variable in the drafted message?

Answer

Yes. If you sequence a prospect and the sequence creates a drafted message that is pending until it is delivered, and you then update any custom fields (on standard objects) that are referenced as variables in the message, the system will automatically update the drafted message for you so it's using the new field values in your variables.
